Job Description

The post-doc will be testing the hypothesis that activation of PKC, Pyk2, TAK1, JAK2, and Src promotes decrease of Cx43 and increase of Cx45 at gap junctions, with corresponding changes in junctional function. More specifically, the post-doc will use HeLa cells stably expressing Cx45 (HeLaCx45; HeLa are Cx deficient) to determine Cx45 cellular localization (immunofluorescence, IMF), protein level (WB), phosphorylation level (IP Cx45; general anti-pS and -pY Ab), co-localization with each kinase (IMF), and GJIC (dye transfer assay - scrape loading) will be characterized in HeLaCx45 cells transfected with Pyk2, constitutively active TAK1 (TAK1-TAB1 fusion protein), constitutively active JAK2 (V617F), or v-Src. The post-doc will also quantify the level of total and active kinase (WB). The post-doc must have knowledge of modern biochemical and spectroscopic techniques to acquire vital, new information to decipher gap junction regulation through the characterization of protein-protein interactions.
